    Qualitative control of Selectivity of liquid medium, mixed culture

    1. Use a quantitative reference material or test suspension with a known number of Cronobacter sakazakii.
    2. Inoculate one tube of test medium with < 100 cells of target microorganisms and with >103 cells of non-target organisms.
    3. Incubate at (41,5 ± 1) °C for (24 ± 2) h.
    4. Remove 10 µl and streak to a plate of specific medium for the target organism.
    5. Incubate at (41,5 ± 1) °C for (24 ± 2) h.
    6. Growth of target microorganisms should reach "Good growth (2)" with a characteristic reaction of: Small to mediumsized (1mm to 3mm) blue to bluegreen colonies.

    Qualitative control of Selectivity solid medium

    1. Use a reference material or test suspension with a known number of Staphylococcus aureus.
    2. Streak the test strain (Staphylococcus aureus) as a single line on the media under test (CCI).
    3. Use a 1 µl loop in case of a concentration of at least 10000 cfu or a 10 µl loop in case of a concentration between 1000 and 10000 cfu.
    4. Incubate at (41,5 ± 1) °C for (24 ± 2) h.
    5. Asses the amount of growth as, 0 for no growth, 1 for weak growth or 2 for good growth.
    6. For this medium (CCI) and standard (ISO 22964) combination the criteria is: "No growth.

    2. Streak the test controlstrain (Enterobacter cloacae) on the media under test (CCI) to obtain discrete colonies.
    3. Use a 1 µl loop in case of a concentration of at least 1000 cfu/mL.
    4. Incubate at (41,5 ± 1) °C for (24 ± 2) h.
    5. Asses the presence, absence and/or grade of expression of biochemical responses and colony sizes and morphology.
    6. For this medium (CCI) and standard (ISO 22964) combination the characteristic reaction is: Colonies do not have a green or bluegreen colour.
