ISO 6579-1 (2017) medium: MSRV agar

Media control Food chain

Microorganisms and performance criteria for selective enrichment media 'MSRV agar' used in Food chain microbiology according to ISO 6579-1.

  • Qualitative control of Productivity

  • Best Practice MSRV agar

Strain free of choice; one of the strains has to be used as a minimum

No further Best Practice available. For the correct method of testing see ISO 11133.

Control strain WDCM number Incubation Criteria Characteristic reaction
Salmonella Typhimurium 41,5 °C ± 1 °C for 2x(24 h ± 3 h) Grey-white, turbid zone extending out from inoculated drop(s). After 24 h to 48 h, the turbid zone(s) will be (almost) fully migrated over the plate. Possible extra: characteristic colonies after subculturing on XLD agar
    Salmonella Enteritidis or

  • 1 Qualitative control of Selectivity

  • Best Practice MSRV agar

Qualitative control of Selectivity of liquid medium, single culture

    1. Use a quantitative reference material or test suspension with a known number of Escherichia coli.
    2. Inoculate one tube of test medium with >103 cells of non-target organisms.
    3. Incubate at 41,5 °C ± 1 °C for 2x(24 h ± 3 h).
    4. Remove 10 µl and streak to a plate of reference medium (TSA).
    5. Incubate at 41,5 °C ± 1 °C for 2x(24 h ± 3 h).
    6. Growth of non-target microorganisms should comply with the criteria: "Possible growth at the place of the inoculated drop(s) without a turbid zone".
